Key Findings
Datroway® (datopotamab deruxtecan), a TROP2-directed DXd antibody-drug conjugate (ADC) jointly developed and commercialized by Daiichi Sankyo and AstraZeneca, has secured approval in China for the treatment of adult patients with unresectable or metastatic triple-negative breast cancer (TNBC). Datroway is the first TROP2-directed ADC to demonstrate an overall survival (OS) benefit in this indication to be approved in China, representing a significant advancement for a highly aggressive and difficult-to-treat form of breast cancer.
Technical / Clinical Details
The approval is underpinned by the groundbreaking results from the Phase 3 TROPION-Breast02 trial, which were presented at the 2025 European Society for Medical Oncology (ESMO) Congress and subsequently published in the Annals of Oncology. The TROPION-Breast02 study evaluated the efficacy and safety of Datroway in patients with previously untreated or pre-treated unresectable or metastatic TNBC. The trial demonstrated a statistically significant and clinically meaningful prolongation of overall survival in the Datroway arm compared to standard chemotherapy. While specific OS duration values have not been publicly disclosed, achieving the ‘overall survival prolongation’ endpoint is considered a paramount achievement in TNBC, where treatment options are scarce. The observed safety profile was manageable, reflecting the high target specificity of TROP2 ADCs and the effective delivery of the cytotoxic payload, which collectively contributed to this landmark outcome.
Background & Context
Triple-negative breast cancer is defined by the absence of estrogen receptor, progesterone receptor, and HER2 expression, rendering it unresponsive to common hormone therapies or HER2-targeted treatments. It is recognized as one of the most aggressive and prognostically unfavorable subtypes of breast cancer. Effective treatment options for advanced TNBC patients are critically limited, making the emergence of a therapy that extends overall survival a major breakthrough in an area of high unmet medical need. Datroway targets the TROP2 protein, which is highly expressed on the surface of cancer cells, delivering a potent topoisomerase I inhibitor payload, DXd, specifically to tumor cells, thereby selectively killing cancer cells. Given China’s large breast cancer patient population, Datroway’s approval holds significant implications for numerous individuals.
Strategic Significance & Outlook
Datroway’s approval in China will further solidify its global standing as a leading TROP2 ADC. This achievement not only validates the efficacy of the TROP2-targeting ADC strategy but is also expected to accelerate the expansion of Datroway’s indications and its development in other solid tumor types. Potential expedited approval processes in other countries and regions are also anticipated, offering new hope to TNBC patients worldwide. This success reiterates the robust partnership between Daiichi Sankyo and AstraZeneca and their leadership in ADC technology, propelling forward the frontiers of cancer treatment.
