Key Findings
At the 17th World ADC Congress in San Diego, VERAXA Biotech presented in vivo proof-of-concept data showing strong tumor regression in a xenograft breast cancer model using a single dose of 3 mg/kg of BiTAC-ADCs. Notably, the treatment arm showed no decrease in body weight, indicating a differentiated safety profile compared to conventional ADCs. In contrast, the administration of only one non-targeted component of the two-component system resulted in no significant effect.
Technical Details
The BiTAC-ADC platform employs a dual-targeting strategy using two complementary precursors to achieve tumor-restricted activation. The system consists of:
- A systemically inactive prodrug.
- A cell-impermeable proactivator.
These components are delivered via two separate antibodies, each targeting a specific tumor-associated antigen. Cytotoxicity is dose-dependent and occurs only upon the selective internalization of both components into the same cancer cell. In vitro data further confirmed that BiTAC-ADCs can discriminate between cancer and healthy cells and do not release active toxins in the periphery.
Background & Context
VERAXA Biotech, founded on research from the European Molecular Biology Laboratory (EMBL), is developing next-generation antibody-based therapeutics. The company’s BiTAC-ADC technology was evaluated across various cancer cell lines and several validated ADC targets. To validate the results, VERAXA used a control ADC with the same drug-antibody ratio (DAR), conjugation, and linker-payload technology as the BiTAC-ADC counterpart.
Significance & Outlook
The validation of the BiTAC-ADC concept establishes a second pillar in VERAXA’s R&D and partnering strategy alongside BiTAC-TCEs. By ensuring that the therapeutic effect is only induced upon the arrival of both BiTAC components at the tumor site, the company aims to provide safer drug options for patients with solid tumors by reducing the systemic toxicity typically associated with traditional ADC therapies.
