Key Findings
Syntax Bio presented research at the ISSCR iPSC 20th Anniversary Symposium in Kyoto, Japan, showing that its Cellgorithm platform can program human iPSCs toward pancreatic beta cells in four days. The company has completed the screening and selection milestones (Aim 1) of its pancreatic beta cell program, supported by funding from Breakthrough T1D. The project is now moving into Aim 2, which focuses on optimization, process development for scalable manufacturing, and preclinical evaluation in collaboration with Mayo Clinic.
Technical Details
The Cellgorithm platform employs CRISPR activation (CRISPRa) to trigger the cell’s own genes without editing or cutting the genome. This process mimics human development by activating genes in a specific sequence and timing via a single delivery of transient DNA instructions. To achieve these results, researchers tested over 170 Cellgorithm variants across three screens, utilizing 29 pancreatic lineage genes. Each variant activated up to 10 genes in five or fewer timed steps.
Analysis via protein expression and single-cell RNA sequencing confirmed that specific cells reached endocrine precursor and pancreatic beta-like states within the four-day window. The research also identified “gridlocks,” where cells stalled at intermediate stages despite receiving instructions for later stages.
Background & Context
The findings were shared during an oral presentation and poster (Poster 288) titled “Cellgorithms: Programming Human iPSC Differentiation With Modular, Time-Resolved CRISPRa Cascades,” delivered by co-founder and CSO Ryan Clarke on October 21, 2026. The platform is designed to replace traditional, labor-intensive differentiation processes that typically take months with a programmable approach that takes days or weeks.
Significance & Outlook
Each program generated by the platform maps the human cell lineage tree, creating compounding data that improves the platform’s ability to predict efficient paths to desired cell states. Syntax Bio intends to apply these insights to every cell type it programs, aiming to create a scalable, reliable manufacturing process for regenerative medicines, specifically targeting transformative treatments for type 1 diabetes.
