Key Findings
UDC-NK cells demonstrated near-complete tumor growth inhibition and significantly prolonged survival in disseminated B-cell lymphoma models when used in combination with Rituximab. In vitro tests showed robust cytotoxicity and cytokine production against CD20⁺ lymphoma cell lines, with efficacy further enhanced by the addition of Rituximab. Additionally, the cells successfully evaded immune responses mediated by allogeneic T cells and NK cells.
Technical Details
The UDC-NK platform utilizes a multi-gene-edited induced pluripotent stem cell (iPSC) line. To achieve immune evasion, the researchers disrupted B2M and RFXANK to eliminate HLA class I and II molecules and introduced a single-chain HLA-E. For safety and controllability, a herpes simplex virus thymidine kinase–based safety switch was incorporated, allowing for the selective elimination of UDC-NK cells via ganciclovir treatment in vitro.
The manufacturing process is characterized by:
- A novel feeder-free, scalable process.
- Production of cryopreserved UDC-NK cells for off-the-shelf application.
- Generation of large quantities of cells exhibiting an activated phenotype.
Background & Context
Natural killer (NK) cells are valued in cancer immunotherapy for their intrinsic ability to eliminate malignant cells and their safety profile. However, clinical application has historically been hindered by donor variability, the risk of immune rejection, and difficulties associated with large-scale manufacturing.
Significance & Outlook
The development of the UDC-NK platform addresses the primary bottlenecks of donor dependency and scalability. By combining immune-evading genetic edits with a scalable manufacturing process and a built-in safety switch, this platform establishes a foundation for off-the-shelf cellular therapies. The results support the potential clinical application of these cells, particularly when used in combination with antibody-based cancer therapies.
