Key Findings
Syntax Bio presented research at the ISSCR iPSC 20th Anniversary Symposium showing that its Cellgorithm platform can program human iPSCs toward pancreatic beta cells in four days. The company has completed the screening and selection milestones (Aim 1) of its pancreatic beta cell program, supported by Breakthrough T1D. The project is now moving into Aim 2, which focuses on optimization, process development for scalable manufacturing, and preclinical evaluation in collaboration with Mayo Clinic.
Technical Details
The Cellgorithm platform employs CRISPR activation (CRISPRa) to trigger the cell’s own genetic instructions without editing or cutting the genome. The system mimics human development by activating genes in a specific sequence and at controlled times via a single delivery of transient DNA instructions. To achieve these results, researchers conducted three screens testing over 170 Cellgorithm variants derived from 29 pancreatic lineage genes. Each variant activated up to 10 genes in five or fewer timed steps.
Analysis via protein expression and single-cell RNA sequencing confirmed that specific cells reached endocrine precursor and pancreatic beta-like states within the four-day window. The research also identified “gridlocks,” where cells stalled at intermediate stages despite the presence of later-stage instructions.
Background & Context
The findings were presented by co-founder and CSO Ryan Clarke on October 21, 2026, in Kyoto, Japan. The Cellgorithm platform is designed to replace traditional, labor-intensive differentiation processes that often take months. By mapping the human cell lineage tree, the platform creates a compounding data set that improves the prediction of efficient paths to desired cell states.
Significance & Outlook
By reducing the time required to generate specialized cells to days or weeks, Syntax Bio aims to overcome bottlenecks in the production of regenerative medicines. The company is currently collaborating with Mayo Clinic to combine the Cellgorithm platform with expertise in pancreatic biology and clinical translation to develop a transformative cell therapy for patients with type 1 diabetes.
